Is there an advantage (Apart from esthetics) in keeping a 5HP outboard on an adjustable transom bracket or placing in outboard well as per design specs for CD22?

Aesthetics aside, keeping the outboard on the transom means no big hole in the hull. You also avoid having to choose between removing and storing the engine each time you tie up for the evening or leaving it in the water to acumulate crud in the cooling system.

In my opionion, an important consideration is how the placement of the motor affects the trim of your boat. Hanging an additional 70 lbs. or so behind a relatively light boat like the 22 probably affects trim as much as adding an additional adult in the rear of the cockpit because of its leverage effect back there. Compare that to placing the weight further forward in a well, and I'm sure there would be a noticeable difference. Another thing to think about is asthetics.
